Use the probe of animal B-ultrasound diagnostic instrument to identify follicles under the fluorescent screen display. Push the puncture needle from the puncture stand along the direction of the puncture line and puncture the follicle. Pedal the low-pressure suction device to suck the oocyte into the oviduct. After in vitro maturation, the oocyte can be fertilized and can also be used for embryo transplantation. In 1993, Boni et al. in Italy first reported the study of live egg collection of buffalo. In 1998, Galli et al. used two dry-milk female buffaloes to collect live oocytes and combined with in vitro fertilization technology to produce in vitro embryos. Then, 9 frozen embryos were thawed and transplanted to 9 recipient cows in estrus. Finally, 3 were confirmed to be pregnant and all developed to the full term of pregnancy. For the first time, 3 surviving buffalo calves were successfully born. The research on live egg collection technology of buffalo in my country started late. Huang Youjun et al. [1] (2004) reported that 42 fine river buffaloes were live egg collection using animal B-ultrasound ultrasound egg collection device. Eggs were collected once every 3 days. Eggs were collected 2-3 times in succession. A total of 96 times of egg collection were performed. 762 follicles were punctured and 513 oocytes were recovered. The recovery rate was 67.3%. Among them, 305 oocytes were usable. The usability rate was 59.5%: an average of 5.34-4.25 oocytes were obtained per time. Among them, 3.18-2.89 oocytes were usable. The usable oocytes were cultured in two systems, cell co-culture and synthetic oviduct fluid culture (SOF). The results showed that the fertilization rate and blastocyst rate were 45.2%, 30.6% and 70.8%, 18.8% respectively. Eight embryos were transplanted into eight swamp-type recipient cows. Finally, two cows were confirmed to be pregnant and all developed to the full term of pregnancy. The world's first river-type buffalo bred by swamp-type buffalo was born. Animal B-ultrasound diagnosis is of great significance to improving the management level of animal husbandry. It is of great significance to promote the economic development of animal husbandry. Using animal B-ultrasound diagnostic technology to evaluate the fertility of buffaloes can help us understand and know the status of buffalo ovaries and uterus during estrus cycle and pregnancy, and better improve reproductive performance and promote animal genetic progress.
